Thin layer chromatography (TLC) analysis and antihyperuricemic activity of bamboo shoots extract {Schizostachyum brachycladum Kurz (Kurz)} in male white mice

Thin-layer chromatography (TLC) analysis of bamboo shoot extract (Schizostachyum brachycladum) and its antihyperuricemic activity evaluation on male mice have been conducted. The extraction was processed by the maceration method, while the determination of the compound was carried out by using various color reagents of TLC plate. To evaluate the antihyperuricemic activity of the extract, a total of 25 male mice were hyperuricemic-induced with daily chicken liver homogenates along with extract suspension at doses of 25, 50 and 100 mg/kg, allopurinol 10 mg/kg, and control group. Another group consisted of 5 mice was treated as the normal group. Serum uric acid level of mice was measured by using the spectrophotometer at the 8th day and analyzed with one-way ANOVA. The TLC analysis showed that the extract contained phenolic compounds and triterpenoids. The measurement of serum uric acid levels revealed that the extract at doses of 25, 50 and 100 mg/kg significantly decreased the uric acid levels of male white mice as compared with control (p<0.05). However, all doses did not show any significant difference in reducing the uric acid level (p>0.05). The study concluded that bamboo shoot extract exhibited antihyperuricemic activity. The effect was seen even at the lowest dose evaluated.

The aim of this study was to know the water fraction effect of Salam Leaves (Syzygium polyanthum Wight.Walp) on rats, which would be focused on the effect in lowering uric acid levels in blood on male hyperuricemic-diabetic rats. Material used in this research were n-hexane fraction, ethyl acetate fraction and water fraction of the leaves. Hyperuricemic rats were obtained by giving 3%b/b cow liver homogenates given orally for 15 days in the dose of 200 mg/kg for each rats. The water fraction was given orally for 15 days in the doses of 50; 200 and 250 mg/kg. The blood uric acid levels were measured by Easy Touch® GCU Multi Functions Monitor System (analyzer). The decreasing data of uric acid levels were analyzed by two-way ANOVA and Duncan’s Multiple Range Test. The result of this study showed that water fraction decreased uric acid levels better than n-hexane and ethyl acetate fraction. Uric acid levels were influenced by the dose and the duration of Salam leaves fraction treatment (p < 0.05). The percentage of uric acid levels decrease was influenced by the interaction between dose and duration of treatment (p < 0.05). The water fraction dose given to hyperuricemic-diabetic rats influenced the percentage decrease of uric acid levels. Pathological condition of male hyperuricemic-diabetic decrease the activity of Salam leaves water fraction to lower uric acid levels, hence the adjustment of doses is needed for hyperuricemic-diabetic rats.
